Abstract
TCF7L2 gene is one of the genes that cause type-2 diabetes mellitus (T2DM). The purpose of the research are: 1. The primary construct for the gene variant rs7895340 TCF7L; 2. Knowing polymorphism ability to detect primary gene TCF7L2 rs7895340 variant. The research method used was a cross-sectional study, the main steps: DNA isolation, primer design for gene TCF7L2 use primary software designer, amplification of the gene TCF7L2 rs7895340 variant with ARMS-PCR method, and the method then direct DNA sequencing bioinformatics analysis. Based on the analysis of data it can be concluded that it has successfully constructed three primary i.e. RS78F forward primer, reverse primer and primer forward RS78R RS78C. The third primary is constructed able to recognize the SNP rs7895340 TCF7L2 gene by ARMS-PCR method.
