Singkong (Manihot esculenta) berpotensi sebagai bahan baku bioetanol karena kandungan patinya tinggi. Penelitian ini bertujuan memproduksi dan mengkarakterisasi bioetanol dari singkong serta menganalisis pengaruh suhu fermentasi terhadap kadar etanol yang dihasilkan. Proses produksi meliputi liquifikasi dengan enzim α-amilase, sakarifikasi dengan glukoamilase, fermentasi menggunakan Saccharomyces cerevisiae selama 7 hari pada suhu 30 °C, 35 °C, dan 40 °C, serta destilasi pada 78 °C. Kadar etanol diukur dengan alkoholmeter dan karakterisasi kimia menggunakan FTIR. Hasil menunjukkan kadar etanol tertinggi diperoleh pada suhu 30 °C sebesar 73% (214 g), diikuti 35 °C sebesar 68% (187 g), dan 40 °C sebesar 63% (170 g). Spektrum FTIR mengonfirmasi keberadaan gugus O–H, C– H, dan C–O khas etanol.
