FOS dan fruktosa merupakan dua senyawa penting di industri pangan. Penelitian ini bertujuan untuk menentukan aktivitas inulinase murni parsial pada variasi pH (3 ; 3,5; 4,0; 4,5; 5,0; 6,0; dan 7,0), suhu (20ºC; 30ºC; 37ºC; 40ºC dan 50,0ºC), dan konsentrasi substrat (1,0% ; 1,25% ; 1,5% ; 1,75% ; 2,0.% 2,25%; 2,5%; dan 3,0% ) dengan waktu inkubasi 30 menit. Selain itu, juga dilakukan penentuan nilai KM dan Vmaks serta massa molekul dari inulinase murni parsial. Aktivitas inulinase ditentukan dengan metoda Nelson-Samogyi pada λmaks 500 nm. Nilai KM dan Vmaks ditentukan melalui persamaan Michaelis-Menten dan persamaan Lineweaver-Burk. Massa molekul ditentukan dengan metoda SDS-PAGE. Aktivitas optimum inulinase yang diperoleh adalah 0.0748 U/mL pada pH 5,0 ; suhu 40ºC dan konsentrasi substrat 2,25%. Nilai KM dan Vmaks inulinase adalah 4.884 % (w/v) dan 22.3x10-2 U/mL.
